Data-driven optimised protein production using Pichia pastoris

Our project pioneers a data-driven approach to improving protein biomanufacturing through predictive gene design. Building on the success of our proprietary machine learning platform, codABLE™, which enhances recombinant protein expression in prokaryotes, we are now extending its capabilities to Pichia pastoris, a key eukaryotic host in industrial biotechnology. By integrating high-throughput fluorescence-activated cell sorting and next-generation sequencing, we generated a large dataset linking gene design to expression outcomes. This dataset was used to train and refine a machine learning model and uncover critical relationships between codon usage and protein production. Unlike other codon optimisation tools, codABLE™ enables precise tuning of gene expression without altering regulatory elements, addressing key challenges in therapeutic protein and industrial enzyme development, and significantly advancing sustainable biomanufacturing practices.
